Radioresistance represents a substantial challenge in cancer treatment. Using Arraystar snoRNA PCR Array, snoRNA SNORA58 was found up-regulated in esophageal squamous cell carcinoma (ESCC). SNORA58 enhances cancer cell resistance to radiotherapy by inhibiting ubiquitin-mediated degradation of CTCF, leading to downregulation and inactivation of JNK signaling pathway, disrupted intracellular iron homeostasis, and radiotherapy-induced ferroptosis. These findings are the first to demonstrate SNORA58 as a critical regulator of cancer radioresistance, suggesting potential therapeutic strategies for ESCC.
