rG4 is self-assembled guanidine quartets via Hoogsteen bonds that stack into an RNA quadruplex. rG4s can dynamically form in the 5’UTR, 3’UTR, and ORF of an mRNA in vivo, as novel RNA elements to regulate protein translation, RNA stability, and epitranscriptomic modifications. rG4s also play diverse roles in non-coding RNAs, e.g. telomere stability, phase separation, and functional regulation of miRNAs, piRNAs, tRNAs, and lncRNAs. rG4s are associated with cancers and diseases, making them an important RNA feature for scientific and clinical research.
