Nature Cancer: tRNA modification reprograms cancer translation and fuels glioblastoma growth

Threonine is a substrate for t6A modification of tRNAs by YRDC enzyme. When accumulated in glioblastoma cells due to altered cancer metabolism, t6A modified tRNAs increase global translation and shift the translated proteome by their changed codon bias toward mitosis-related genes [1].

Arraystar tRNA-seq was used to analyze tRNA isoform abundances under the condition. The study elucidates the mechanism of translational and metabolic reprogramming in cancer. It provides a molecular basis for dietary threonine restriction to reduce tumor t6A formation, inhibit tumor growth, and augment cancer chemotherapy efficacy.
