circPTEN circular RNA was discovered down regulated in clear cell renal cell carcinoma (ccRCC) on Arraystar circRNA Microarray [1]. circPTEN regulates DNMT1 which epigenetically controls tumor suppressor PTEN gene expression (Fig. 1, left), and METTL3 which epitranscriptomically modifies glucose transporter GLUT1 mRNA (Fig. 2, right). Both mechanisms suppress the kidney cancer and mTOR inhibitor cancer drug resistance via mTOR pathway.
