Recently published in Mol. Cell (2022), a lncRNA called glycoLINC (gLINC) was discovered on Arraystar LncRNA microarray as the top lncRNA in c-Myc-mediated regulation of cell metabolism. gLINC acts as a backbone to assemble glycolysis enzymes (PGK1, PGAM1, ENO1, PKM2, and LDHA) into the metabolon (Fig.1)[1], which greatly increases glycolytic flux, ATP production, and cell survival under serine deprivation. Furthermore, gLINC overexpression reprograms the cancer cell metabolism to promote xenograft growth in mice on serine deficient diet. gLINC is the first example of lncRNA-assembled metabolon in the glycolysis pathway.
